Product Specific Information

This 22F6 monoclonal antibody reacts with human, mouse, and canine Helios. Helios is a zinc-finger transcription factor related to Ikaros, expressed in hematopoietic stem cells and highly in regulatory T cells, but not in mature B cells, dendritic cells, or myeloid cells. Helios can form homo- or heterodimers with other Ikaros family members to regulate transcription. Eight isoforms exist, some with dominant-negative functions linked to lymphoid malignancies such as T-acute lymphoblastic leukemia.

22F6 has been reported to cross-react with rat Helios.

Applications Reported

• Intracellular staining followed by flow cytometric analysis.

Applications Tested

• Pre-diluted and tested by intracellular staining of mouse lymph node cells using the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00).
• Recommended use: 5 µL (0.125 µg) per test (100 µL final volume).
  Cell count: 10⁵–10⁸ cells/test (empirically determined).

Brilliant Violet™ 480 (BV480) emits at 479 nm and is suitable for cytometers with a 405 nm violet laser. Ensure compatibility of your instrument.

When using multiple polymer dye–conjugated antibodies (Super Bright, Brilliant Violet™, Brilliant Ultra Violet™), use Super Bright Complete Staining Buffer (Product #SB-4401-42) or Brilliant Stain Buffer™ (Product #00-4409-75) to minimize non-specific interactions.

Note: Brilliant Violet™ 480 (BV480) is not compatible with methanol-based fixation.

Target Information

This gene encodes a member of the Ikaros family of zinc-finger proteins (Ikaros, Aiolos, and Helios). These transcription factors regulate lymphocyte development and function primarily in early hematopoietic development. Multiple transcript variants exist, though not all have confirmed biological validity.
