Target Information

Trex1 (ATRIP) is the major human 3′ to 5′ exonuclease required for checkpoint signaling after DNA damage. It binds to single-stranded DNA coated with replication protein A, recruiting ATR kinase to sites of DNA damage and replication stress. Trex1 is essential for ATR expression.
This gene utilizes two different open reading frames:

• The upstream ORF encodes proteins interacting with ATR and localizing to intranuclear foci after DNA damage.
• The downstream ORF encodes exonuclease-active proteins, possibly a subunit of human DNA polymerase III.

Mutations in TREX1 are associated with Aicardi-Goutieres syndrome, chilblain lupus, and Cree encephalitis.
