Product Specific Information
PA1-013 detects KDEL from human, rat, mouse, and hamster.
This antibody has been successfully used in Western blot, immunofluorescence, and immunoprecipitation procedures.
- Western Blot Results: Detects a 57 kDa protein (PDI), a 78 kDa protein (GRP78), and an ~94 kDa protein (GRP94) in mouse myeloma cells.
- Immunofluorescence: Staining of GRP78 in mouse myeloma cells shows diffuse cytoplasmic staining consistent with ER localization.
- Immunoprecipitation: PA1-013 immunoprecipitates PDI, GRP78, an ~72 kDa ERp72 protein, and GRP94 from rodent tissues.
Reconstitution: Reconstitute in 100 µL PBS to create a stock of 1 mg/mL.
Target Information
The sequence Lys-Asp-Glu-Leu (KDEL) or closely related motifs are found at the C-terminus of soluble ER-resident and some membrane proteins.
GRP78 (78 kDa), GRP94 (94 kDa), and PDI share this C-terminal KDEL sequence.
This motif is essential for ER retention and reduces secretion of proteins from the ER, mediated by the KDEL receptor.
