Product Specific Information

PA1-012 detects phospho-alpha-B crystallin (Ser59) from bovine, human, and rat samples.
This antibody does not detect the unphosphorylated form of the protein.

• Validated for Western blot, immunohistochemistry, and immunofluorescence.
• Detects an ~20 kDa recombinant bovine phospho-alpha-B crystallin (Ser59) by Western blot.
• In immunofluorescence staining of human brain tissue from Alexander’s disease patients, positive staining is observed in cells of the cerebral cortex.
• Immunogen: Synthetic phosphopeptide corresponding to residues F(54) L R A P S(p) W I D T G(64) of human alpha-B Crystallin.
• Reconstitute in 100 µL PBS to create a 1 mg/mL stock solution.

Target Information

Lens proteins consist predominantly of crystallins (~95%), which are also found in vertebrate skeletal muscle tissue.
In the lens, crystallins maintain the proper refractive index and structural integrity.

• Major classes: alpha, beta, and gamma crystallins.
• Alpha-crystallin (comprising alpha-A and alpha-B) constitutes about 40% of total lens protein.
• Functions as a molecular chaperone preventing aggregation and potential cataract formation.
• Phosphorylation of alpha-crystallin occurs in response to cellular stress, potentially regulating protein maintenance.
• Alpha-B crystallin accumulation is associated with Alexander’s disease in human brain cells.
