Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nucleotide deprivation. Upon activation, p53 induces transcription of genes such as p21waf1, leading to G1 or G2/M arrest.

Structurally, p53 contains:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerization domain
  • C-terminal regulatory domain

Phosphorylation at Ser15 is critical for p53 activation and stabilization, while phosphorylation at Ser392 is associated with tumor formation. p53 plays a key role in preventing proliferation of damaged cells, acting as an anticancer agent. Elevated p53 levels have been linked to aging, oxidative stress, and impaired long-term potentiation (LTP) in learning and memory.

In normal cells, p53 is expressed at low levels, but in many transformed cell lines, it is highly expressed and contributes to malignancy. Mutations in the TP53 gene disrupt DNA binding and tumor suppressor activity, and such mutations are common in human cancers and familial cancer syndromes like Li-Fraumeni syndrome.
