Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or pyrimidine nucleotide starvation. Upon stress, p53 up-regulates and activates transcription of genes such as p21waf1, leading to G1 or G2/M arrest.

Structurally, p53 contains:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerisation domain
  • C-terminal regulatory domain

Phosphorylation at Ser15 is critical for activation and stabilization, while phosphorylation at Ser392 is associated with tumor formation. p53 plays a key role in preventing proliferation of damaged cells and functions as an anticancer agent. It is expressed at low levels in normal cells but highly in transformed cell lines, contributing to malignancy. Mutations in the TP53 gene result in loss of tumor suppressor activity and are common in various human cancers, including germline mutations seen in Li-Fraumeni syndrome.
