Product Specific Information

  • For immunohistochemistry (PFA-fixed): Incubate antibody for 30 minutes at room temperature.
  • Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation. Upon activation, p53 upregulates genes such as p21^WAF1^, leading to G1 or G2/M arrest.

The p53 structure includes:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerization domain
  • C-terminal regulatory domain

Phosphorylation at Ser15 is critical for activation and stabilization, while phosphorylation at Ser392 is associated with tumor formation. p53 plays a key role in preventing proliferation of damaged cells and has been linked to cancer suppression, aging, and oxidative stress responses. Mutations in the TP53 gene are common in human cancers and lead to loss of tumor suppressor function. Germline TP53 mutations are associated with Li-Fraumeni syndrome.
