Product Specific Information

This 5H12 monoclonal antibody reacts with mouse Autoimmune Regulator (AIRE). The 58-kDa transcription factor is highly expressed within the perinuclear region of mature medullary thymic epithelial cells (mTECs). Expression of AIRE protein in peripheral tissues remains unclear despite detection of AIRE mRNA in lymph nodes, stromal cells, monocytes, and dendritic cells.
AIRE is involved in thymocyte development and regulation of tissue-specific antigen expression during negative selection, as well as chemokine expression and antigen presentation.
The 5H12 antibody recognizes mouse AIRE isoforms 1 and 2.

Applications Reported: Flow cytometric analysis and immunohistochemical staining of frozen tissue sections (IHC-F).
Applications Tested: Tested by IHC on fixed, frozen mouse thymus at ≤10 µg/mL. Optimal titration recommended.
Excitation: 488 nm; Emission: 519 nm; Laser: Blue Laser.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

AIRE is a transcriptional regulator protein that binds DNA as a dimer or tetramer, not as a monomer, and regulates gene expression. It is a nuclear protein with HSR, two PHD-type zinc fingers, and a SAND domain, forming high-molecular-weight complexes.
AIRE interacts with CBP and CREBBP, and has three isoforms, one potentially subject to nuclear-mediated decay (NMD).
Defects in AIRE cause autoimmune polyendocrinopathy-candidiasis-ectodermal dystrophy (APECED), a rare autosomal recessive autoimmune disease.
AIRE also mediates E3 ubiquitin ligase activity and plays a key role in immune regulation.
