Target Information
Spi-B is a transcription factor belonging to the ETS family with a conserved ETS domain that mediates specific DNA binding.
It interacts with IRF4, CREBBP, EP300, SPI1/PU.1-related proteins, JUN, and TBP. As a sequence-specific transcriptional activator, Spi-B binds to the PU-box (5'-GAGGAA-3'), acting as a lymphoid-specific enhancer.
Spi-B is expressed in plasmacytoid dendritic cells (pDCs) and promotes their development while blocking viral replication.
It is not expressed in T-cells or granulocytes but may be found in liver stem cells and B-cells, aiding B-cell receptor signaling and normal B-cell development.
