The 188 monoclonal antibody was generated against and reacts with mouse B7-H4 (also known as B7S1, B7X). Cross-reactivity to other proteins has not been determined.
B7-H4 is a member of the B7 family that inhibits T-cell activation, cell cycle progression, and IL-2 production.
Double staining experiments using clones 9 and 188 indicate distinct epitopes. Clone 188 stains mouse B7-H4 transfected cells but not spleen cells.
Applications reported: Flow cytometric analysis of transfected cells.
Applications tested: ≤0.5 µg/test (100 µL final volume, 10⁵–10⁸ cells/test).
Careful titration is recommended for optimal assay performance.
Target Information
B7-H4 (B7x, VTCN1) is a costimulatory protein acting as a negative regulator of T-cell mediated immunity.
It delivers inhibitory signals to T-cells, preventing proliferation and IL-2 production.
B7-H4-deficient mice show minimal immune impact, indicating its role in fine-tuning immune responses.
Expressed on activated T-cells, B-cells, monocytes, and dendritic cells.
Aberrant expression is linked to lung, breast, and ovarian cancers.
Overexpression may suppress T-cell immunity and promote tumor progression.
VTCN1 is a potential biomarker for early detection of ovarian cancer and renal cell carcinoma (RCC).
