This antibody detects endogenous levels of HPK1 only when phosphorylated at Ser421.
Target Information
MAP4K1 (also known as HPK1, Hematopoietic progenitor kinase 1) is a hematopoietic cell-restricted member of the Ste20 serine/threonine kinase superfamily.
It acts as a tissue-specific upstream activator of the MEKK/JNK/SAPK signaling pathway and diminishes T cell receptor (TCR) signaling activity and T cell proliferation by phosphorylating the adaptor protein SLP-76.
HPK1 specifically activates JNK1 and enhances AP-1-mediated transcriptional activity. Unlike PAK65, HPK1 does not contain the small GTPase Rac1/Cdc42-binding domain and does not bind to Rac1 or Cdc42, suggesting a distinct regulatory mechanism.
