Product Specific Information

The eBioCB16 monoclonal antibody recognizes CD16 (Fc gammaRIII), a low-affinity receptor for IgG with an apparent molecular weight of 50–80 kDa. CD16 is encoded by two related genes:

  • CD16A (FcγRIIIA): Expressed on macrophages and NK cells (transmembrane form)
  • CD16B (FcγRIIIB): Expressed on neutrophils (GPI-anchored form)

Two polymorphisms, NA-1 and NA-2, exist for CD16B; NA-2 homozygotes show reduced phagocytic capacity. CD16 binds immune complexes of IgG, preferentially IgG1 and IgG3, and mediates ADCC, phagocytosis, degranulation, and proliferation upon activation.

Each product contains:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported

  • Suitable for flow cytometric analysis of human peripheral blood cells.

Applications Tested

  • Pre-diluted and validated at 5 µL (0.2 µg) per test (100 µL final volume).
  • Cell number range: 10⁵–10⁸ cells/test.

Compatibility Information

  • NovaFluor dyes are not compatible with DNA-intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer

  • Updated conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01).
  • Required for optimal staining with NovaFluor conjugates.
  • Add 5 µL per sample to antibody cocktails before cell addition.
  • For single-color controls: 5 µL per 100 µL of sample (10³–10⁸ cells).

Target Information

CD16 (FcγRIII) encodes a receptor for the Fc portion of IgG, involved in:

  • Clearance of immune complexes
  • Antibody-dependent cytotoxicity
  • Enhancement of viral infection responses

The FCGR3A gene encodes the NK cell–associated transmembrane receptor, while FCGR3B encodes the neutrophil-specific GPI-linked receptor.
Mutations are linked to Immunodeficiency 20, Herpes Zoster, and Systemic Lupus Erythematosus susceptibility.
Alternative splicing generates multiple isoforms.
