Product Specific Information

The eBioCB16 monoclonal antibody recognizes CD16 (Fc gammaRIII), a low-affinity IgG receptor (50–80 kDa).
CD16A is expressed in macrophages and NK cells, while CD16B is found in neutrophils with two polymorphisms (NA-1, NA-2).
CD16 mediates immune complex binding and triggers ADCC, phagocytosis, degranulation, and proliferation upon IgG binding.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells at 5 µL (0.6 µg)/test (100 µL final volume; 10⁵–10⁸ cells/test)

NovaFluor Compatibility Notes:

• Not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI)
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):

• Required for optimal staining with NovaFluor conjugates
• Add 5 µL per sample before mixing with cells
• For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells)

NovaFluor Technology:

Target Information

CD16 (FCGR3A) encodes a receptor that binds the Fc portion of IgG, involved in immune complex clearance, ADCC, and modulation of viral infection susceptibility.
FCGR3A is expressed on NK cells as a transmembrane glycoprotein, while FCGR3B (CD16B) is expressed on neutrophils with PI linkage.
Mutations in FCGR3A are linked to Immunodeficiency 20, Herpes Zoster, and autoimmune susceptibility.
