Product Specific Information

The eBioCB16 monoclonal antibody recognizes CD16 (Fc gammaRIII), a low-affinity receptor for IgG (50–80 kDa).
CD16 includes two similar genes:

• CD16A (FcγRIIIA): expressed in macrophages and NK cells
• CD16B (FcγRIIIB): expressed in neutrophils (GPI-anchored form)

Polymorphisms NA-1 and NA-2 affect phagocytic capacity. CD16 binds IgG immune complexes, preferentially IgG1 and IgG3, and initiates signaling cascades leading to ADCC, phagocytosis, degranulation, and proliferation.

• 1 vial of CellBlox Plus Blocking Buffer

Applications Tested: 5 µL (0.6 µg)/test for human peripheral blood cells (10⁵–10⁸ cells/test)

NovaFluor dyes are not compatible with DNA intercalating viability dyes (PI, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for compatibility.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates.
Add 5 µL per sample before combining with cells. For single-color controls, use 5 µL per 100 µL cell sample.

NovaFluor conjugates utilize Phiton™ technology, based on novel nucleic acid dye structures engineered for optimized fluorescence with minimal spillover and spread.

Target Information

CD16 (FCGR3A) encodes a receptor that binds the Fc portion of IgG, mediating immune complex clearance and antibody-dependent cytotoxicity.

• Expressed on NK cells (transmembrane form)
• FCGR3B expressed on neutrophils (PI-anchored form)
  Mutations are associated with Immunodeficiency 20, Herpes Zoster, and Systemic Lupus Erythematosus.
  Alternative splicing yields multiple isoforms.
