Product Specific Information

The eBioCB16 monoclonal antibody recognizes CD16 (Fc gammaRIII), a low-affinity receptor for IgG (50–80 kDa). CD16 includes two genes:

• CD16A (FcγRIIIA): Found in macrophages and NK cells (polypeptide-anchored form)
• CD16B (FcγRIIIB): Found in neutrophils (GPI-anchored form)

Polymorphisms NA-1 and NA-2 affect phagocytic capacity, with NA-2 showing reduced activity. CD16 binds IgG immune complexes, preferentially IgG1 and IgG3, initiating signal cascades for ADCC, phagocytosis, degranulation, and proliferation.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on human peripheral blood cells
Recommended usage: 5 µL (0.6 µg) per test in 100 µL total volume (10⁵–10⁸ cells/test).

Important Note:
NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer:
Updated buffer (Cat. No. C001T06F01) required for optimal staining with NovaFluor conjugates.

• Add 5 µL per sample before mixing with cells.
• For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates use Phiton™ technology with engineered fluorescent signatures designed to minimize spillover and spread.

Target Information

CD16 encodes a receptor for the Fc portion of IgG, mediating immune complex clearance and functions such as cytotoxicity and viral infection enhancement.

• FCGR3A: Expressed on NK cells (membrane-anchored)
• FCGR3B: Expressed on neutrophils (PI-anchored)

Mutations may cause Immunodeficiency 20, Herpes Zoster susceptibility, and Systemic Lupus Erythematosus. Alternative splicing generates multiple isoforms.
