Product Specific Information
This MR9-4 monoclonal antibody reacts with the mouse T cell receptor (TCR) V beta 5.1/5.2 chain. The MR9-4 antibody recognizes the V beta 5.1 and 5.2 chains on T cells from mouse strains with the b haplotype of the Tcrb gene (e.g., C57BL/6, B10, C58/J).
V beta 5.1 and 5.2 are deleted in strains expressing MHC Class II I-E (e.g., Balb/c, C3H, DBA/2). Reports indicate that V beta 5+ T cells recognize the OVA peptide.
Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes at ≤0.25 µg/test (in 100 µL final volume). Optimal titration recommended.
Fluorochrome Information:
PerCP-eFluor® 710 can replace PE-Cy5, PE-Cy5.5, or PerCP-Cy5.5.

- Recommended filters: 685 LP dichroic mirror and 710/40 band pass filter (695/40 acceptable alternative)
PerCP-eFluor® 710 conjugates are stable after overnight fixation in 2% formaldehyde at 4°C.

Target Information
T cell receptors (TCRs) are essential for distinguishing self from foreign peptides presented by MHC class II molecules. Abnormal TCR recognition is linked to autoimmune diseases and tumors. Superantigen engagement of TCRs can cause toxic shock syndrome. Autoantibodies to V beta segments have been identified in rheumatoid arthritis (RA) and systemic lupus erythematosus (SLE), potentially modulating immune responses.
Most human T cells express TCR αβ with CD4 or CD8 markers, while a small population (DN T cells) lacks both. γδ T cells, found mainly in epithelia, exhibit limited diversity and distinct antigen recognition, contributing to antitumor and immunoregulatory functions.
