Product Specific Information

Description:
The H57-597 monoclonal antibody reacts with the beta chain of mouse TCR. TCR beta is expressed by thymocytes in a developmentally regulated manner and by most peripheral T cells. Crosslinking of the TCR complex with H57-597 induces activation and proliferation of T cells or apoptosis depending on assay conditions. This antibody is used as a phenotypic marker for TCR beta-expressing T cells.

Reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤ 0.25 µg per test.
A test is defined as the amount of antibody that will stain a cell sample in a final volume of 100 µL.
Cell number should be determined empirically (10⁵–10⁸ cells/test).
Antibody titration is recommended for optimal performance.

Fluorophore Information:
eFluor® 450 is an alternative to Pacific Blue®, emits at 445 nm, and is excited with the violet laser (405 nm). Ensure that the instrument can detect this fluorochrome.

Target Information

T cell receptors (TCRs) discriminate foreign from self-peptides presented by MHC class II molecules, which is essential for adaptive immune responses. Recognition of self-peptides is linked to autoimmune diseases, while mutant self-peptides are associated with tumors. Engagement of TCRs by bacterial superantigens can cause toxic shock syndrome.
Autoantibodies to V beta segments of TCRs have been found in patients with rheumatoid arthritis and systemic lupus erythematosus, potentially modulating inflammatory responses.
Human T cells typically express TCR alpha-beta along with CD4 or CD8, while gamma-delta T cells exhibit distinct antigen recognition and are found primarily in epithelial tissues.
