• Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature (RT).
• For formalin-fixed tissues: Heat tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, then cool at RT for 20 min.
• This antibody shows broad species reactivity.

Target Information

PCNA (polymerase delta auxiliary protein) is essential for DNA replication and is involved in DNA excision and mismatch repair pathways.
PCNA binds to CDK inhibitor p21, structure-specific endonucleases Fen1 and XPG, and DNA cytosine 5-methyltransferase (MCMT).
It serves as a marker for proliferating cells and is useful for assessing tumor proliferation status.
Located in the nucleus, PCNA acts as a cofactor of DNA polymerase delta, functioning as a homotrimer to enhance leading strand synthesis during replication.
Upon DNA damage, PCNA becomes ubiquitinated and participates in the RAD6-dependent DNA repair pathway.
Two transcript variants encode the same protein, and pseudogenes have been described on chromosome 4 and the X chromosome.
