Product Specific Information

  • Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature (RT).
  • For formalin-fixed tissues: Heat tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, then cool at RT for 20 minutes.
  • This antibody shows broad species reactivity.

Target Information

PCNA (Proliferating Cell Nuclear Antigen) is essential for DNA replication and is involved in DNA excision and mismatch repair pathways.
It binds to CDK inhibitor p21, endonucleases Fen1 and XPG, and DNA cytosine 5-methyltransferase (MCMT).
PCNA serves as a marker for proliferating cells and is useful for assessing tumor proliferation status.
It is localized in the nucleus, acts as a cofactor of DNA polymerase delta, and forms a homotrimer to enhance leading strand synthesis.
Upon DNA damage, PCNA is ubiquitinated and participates in the RAD6-dependent DNA repair pathway.
Two transcript variants encoding the same protein have been identified, and pseudogenes exist on chromosome 4 and the X chromosome.
