Product Specific Information
- Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
- Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 minutes.
- This antibody shows broad species reactivity.
Target Information
PCNA (polymerase delta auxiliary protein) is essential for DNA replication and is involved in DNA excision and mismatch repair pathways.
It binds to CDK inhibitor p21, endonucleases Fen1 and XPG, and DNA cytosine 5-methyltransferase (MCMT).
PCNA serves as a marker for proliferating cells and tumor prognosis, being expressed in early G1 and S phases.
Located in the nucleus, PCNA acts as a cofactor of DNA polymerase delta, forming a homotrimer to enhance leading strand synthesis.
Upon DNA damage, PCNA is ubiquitinated and participates in the RAD6-dependent repair pathway.
Two transcript variants encoding the same protein have been identified, with pseudogenes on chromosome 4 and X chromosome.
