Product Specific Information
Mature bovine IFNγ was expressed in Pichia pastoris X-33.
GenBank® Accession # AAB64477, amino acids 24–166.
N-terminal sequencing confirmed the first 10 amino acids as QGQFFREIEN.
Purity is >95% by SDS-PAGE analysis.
Activity:
The biological activity of recombinant bovine IFNγ was determined in a viral challenge assay using VS virus on MDBK cells. Plates were stained with crystal violet, visual CPE determination was performed, and absorbance was measured after dye solubilization.
Uses:
For in vitro applications. A dose-response assay is recommended to determine optimal concentration for specific experiments.
Reconstitution:
Reconstitute lyophilized material with 0.5 mL of distilled or deionized water.
Dilute in a buffer containing carrier protein, aliquot, and store at ≤ -70°C.
Avoid multiple freeze-thaw cycles.
PBS with 1% BSA or neutral media containing ≥10% FCS are suitable buffers.
Target Information
Interferon gamma (IFNγ, Type II interferon) is a macrophage activation factor produced primarily by T-lymphocytes and natural killer cells in response to antigens, mitogens, and cytokines.
It is a dimeric glycoprotein (~45 kDa) consisting of two 146 amino acid subunits, showing species-specific biological activity.
Functions include:
- Antiviral and antitumor activity
- Induction of MHC class I and II molecules
- Macrophage activation
- Enhancement of immunoglobulin secretion by B lymphocytes
- Regulation of cytokine signaling via JAK-STAT pathway
Human IFNγ shows ~40% sequence homology with mouse IFNγ and no homology with IFNα or IFNβ.
Expression is upregulated by IL-2, FGF-basic, and EGF, and downregulated by vitamin D3 or DMN.
Mutations in the IFNγ gene are associated with aplastic anemia.
