Product Specific Information

The 2B11 monoclonal antibody reacts with mouse and human CXCR4, also known as CD184, Fusin, LESTR, or HUMSTR. CXCR4 is a G-protein-coupled chemokine receptor that functions as a coreceptor for X4 HIV-1 entry into CD4+ cells and plays a role in lymphocyte migration. Its ligand is SDF1.

CXCR4 is essential for B cell development and maintenance of granulocyte progenitors in bone marrow. Expression patterns differ between mouse and human: restricted to CD4+CD8+ thymocytes in mice and predominantly on B lymphocytes in spleen.

The 2B11 antibody recognizes the amino terminus of CXCR4, induces receptor internalization (without blocking SDF1a binding), and is suitable for flow cytometric analysis of mouse thymocytes. Recommended usage ≤0.5 µg/test (100 µL final volume).

Target Information

CXCR4 (Fusin, LESTR, HUMSTR) is a member of the G-protein-coupled chemokine receptor family with seven transmembrane domains. It functions as a co-receptor for HIV-1 and HIV-2 entry via gp120-CD4 interaction, and mediates Env-induced fusion. CXCR4 binds CXCL12/SDF-1, increasing intracellular Ca²⁺ and activating MAPK1/MAPK3 pathways.

It also acts as a receptor for extracellular ubiquitin, influencing calcium and cAMP levels. CXCR4 is involved in hematopoiesis, cardiac septum formation, and vascular development. In the CNS, it supports neuronal survival and cerebellar development. CXCR4 binding to bacterial LPS triggers inflammatory responses, including TNF secretion.

Antibodies against CXCR4 can block HIV-1 and HIV-2 fusion and infection. The amino-terminal domain and second extracellular loop serve as HIV binding sites.
