Description:
The dG9 antibody clone reacts with human perforin (pore-forming protein, pfp). Perforin is a cytolytic mediator present in the cytoplasmic granules of cytotoxic T lymphocytes (CTL) and natural killer (NK) cells. It plays a critical role in immune responses against tumors and viral infections.

Applications Reported:
This antibody is suitable for intracellular staining followed by flow cytometric analysis.

Applications Tested:
Pre-titrated and validated for intracellular staining and flow cytometric analysis of human peripheral blood cells. Recommended use: 5 µL (0.06 µg) per test (in 100 µL final volume). Suitable for 10⁵–10⁸ cells/test.

Target Information

Perforin is a major cytolytic protein stored in cytoplasmic granules of cytotoxic lymphocytes. It forms transmembrane pores, similar to complement component C9, and is essential for T-cell and NK-cell mediated cytolysis.
Defects in the perforin gene are associated with familial hemophagocytic lymphohistiocytosis type 2 (HPLH2), a rare autosomal recessive disorder.
Perforin protein: 555 amino acids, 21 amino acid signal peptide, molecular weight 70–75 kDa.
Expression is restricted to killer cell lines, not helper T cells or other tumor cells.
