Target Information
LSD1 is a histone demethylase that specifically demethylates Lys-4 of histone H3, a mark associated with epigenetic transcriptional activation. It functions as a corepressor and contains a SWIRM domain, a FAD-binding motif, and an amine oxidase domain.
LSD1 is a component of histone deacetylase complexes and silences genes via demethylation. It oxidizes substrates through FAD to generate imine intermediates that are hydrolyzed.
This enzyme demethylates mono- and tri-methylated Lys-4 of histone H3 and may also act on Lys-9, influencing androgen receptor target gene expression.
Mutations in LSD1 can cause cleft palate, psychomotor retardation, and distinctive facial features.
