Target Information
LSD1 is a histone demethylase that specifically demethylates Lys-4 of histone H3, a marker of transcriptional activation, thereby acting as a corepressor. It contains a SWIRM domain, a FAD-binding motif, and an amine oxidase domain. LSD1 functions within histone deacetylase complexes and silences genes by demethylation. The enzyme oxidizes substrates via FAD to form imines that are subsequently hydrolyzed. It demethylates mono- and tri-methylated Lys-4 of histone H3 and may also act on Lys-9, influencing androgen receptor target gene expression. Mutations in this gene are associated with developmental disorders such as cleft palate and psychomotor retardation.
