Target Information

Apoptosis plays a major role in normal organism development, tissue homeostasis, and removal of damaged cells. Disruption of this process has been implicated in various diseases such as cancer. Members of the Bcl-2 family are critical regulators of apoptosis, characterized by conserved Bcl-2 homology (BH) domains.
Bcl-rambo, a novel and widely expressed member, localizes to mitochondria and induces apoptosis when overexpressed. This effect can be inhibited by co-expression of inhibitor of apoptosis proteins (IAPs) such as XIAP, cIAP1, and cIAP2.
Bcl-rambo shares homology with anti-apoptotic Bcl-2 family members but has a unique 250 amino acid insertion preceding its C-terminal membrane anchor. This region alone can induce apoptosis more efficiently than the BH domains, suggesting Bcl-rambo's role in additional pro-apoptotic pathways.
