Product Specific Information

The 5C3 monoclonal antibody reacts with human CD40, a 45–50 kDa type I transmembrane glycoprotein of the TNFR family. CD40 is expressed by B lymphocytes, follicular dendritic cells, thymic epithelium, and a subset of peripheral T cells. It regulates B cell development and maturation by inducing Ig isotype-switching and, in combination with IL-4, protects B cells from apoptosis while promoting proliferation. Interaction of CD40 with CD154 (gp39) on T cells is critical for T-B cell crosstalk and immune regulation. The 5C3 clone is used for activation of B cells in functional assays.

Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and validated for flow cytometry of normal human peripheral blood cells at 5 µL (0.6 µg) per test.

A test is defined as the antibody amount required to stain a cell sample in 100 µL final volume. Cell number may range from 10⁵ to 10⁸ cells/test.

Note: NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which improves staining performance. Add 5 µL per sample prior to combining with cells. For single-color controls, use 5 µL per 100 µL of cell sample containing 10³–10⁸ cells.

NovaFluor conjugates utilize Phiton™ technology, incorporating novel nucleic acid dye structures to optimize fluorescence performance and minimize spillover.

Target Information

CD40 is a single-chain glycoprotein of the TNF/NGF receptor superfamily. It is expressed on all B cells except plasma cells, as well as on some epithelial cells, carcinomas, and dendritic cells. CD40 is essential for immune regulation, including T cell-dependent immunoglobulin class switching, memory B cell formation, and germinal center development. Dysregulation of CD40 is linked to Type 3 Hyper-IgM immunodeficiency and CD40 ligand deficiency. It also plays a role in amyloid-beta-induced microglial activation, potentially contributing to Alzheimer’s disease pathogenesis.
