Product Specific Information

Description:
The OX8 monoclonal antibody reacts with rat CD8 alpha, a member of the immunoglobulin superfamily. CD8 alpha heterodimerizes with CD8 beta to form the CD8 antigen expressed on CD4+CD8+ thymocytes, most MHC class I-restricted peripheral T cells, and some macrophages. CD8 binds to MHC class I on antigen-presenting cells during antigen presentation and participates in T-cell receptor signal transduction via Lck kinase association.

Applications Reported:
This antibody is reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of rat splenocytes. Recommended ≤1.0 µg/test (defined as the amount of antibody that stains cells in 100 µL volume). Cell number may range from 10⁵ to 10⁸ cells/test. Optimal titration is recommended.

Fluorochrome Information:
Super Bright 436 can be excited with a 405 nm violet laser and emits at 436 nm. Use a 450/50 bandpass filter or equivalent. Ensure instrument compatibility.

Staining Recommendation:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Target Information

Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family involved in signal transduction, T cell differentiation, and activation. CD8 is expressed mainly on T cells as a CD8α/CD8β heterodimer, acting as a co-receptor for MHC class I molecules. CD8α is essential for MHC-I binding and is also expressed on subsets of T cells, NK cells, monocytes, and dendritic cells as CD8α homodimers. Interaction with MHC-I/peptide complexes triggers Lck kinase recruitment, leading to cytokine production, motility, and CTL activation. Activated CTLs play a key role in pathogen and tumor clearance. Differentiation of naïve CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
