Product Specific Information

The RPA-T8 monoclonal antibody reacts with the human CD8a molecule, a 32–34 kDa cell surface receptor expressed as a heterodimer (CD8α/β) or homodimer (CD8α/α). CD8a is found on thymocytes and subsets of mature T cells and NK cells. It binds to MHC class I and associates with protein tyrosine kinase p56lck, contributing to T-cell development and activation.

• Applications Reported: Flow cytometric analysis
• Applications Tested: Pre-titrated and validated for flow cytometry using normal human peripheral blood cells. Use at 5 µL (0.25 µg) per test (100 µL final volume).
• Excitation/Emission: 405 nm / 436 nm (Violet laser)
• Recommended Filter: 450/50 bandpass or equivalent

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Target Information

Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein involved in signal transduction, T-cell differentiation, and activation. It functions as a co-receptor for MHC class I molecules, facilitating cytotoxic T lymphocyte (CTL) activation. CD8 is expressed on T cells, NK cells, monocytes, and dendritic cells, either as heterodimers (CD8α/β) or homodimers (CD8α/α). Interaction with MHC-I/peptide complexes triggers Lck activation, leading to cytokine production and immune response. Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
