Product Specific Information

Description:
The eBio124-1D1 monoclonal antibody reacts with human CD7 (gp40, Leu9), a 40 kDa receptor and member of the immunoglobulin gene superfamily. The N-terminal region (aa1–107) is highly homologous to Ig kappa light chain, while the C-terminal region is proline-rich, forming a stalk-like structure.

CD7 is expressed on most immature and mature T lymphocytes, T cell leukemias, NK cells, a small subset of normal B cells, and malignant B cells. Cross-linking CD7 modulates T cell and NK cell activity, affecting calcium flux, adhesion molecule expression, cytokine secretion, and proliferation. It directly associates with phosphoinositol 3'-kinase, inducing D-3 phosphoinositide production and tyrosine phosphorylation.

A subpopulation of human CD34(+) CD38(-) cord blood cells expressing CD45RA, HLA-DR, and high CD7 levels shows lymphopoietic potential, generating B cells, NK cells, and dendritic cells but not myeloid or erythroid cells. CD7(+) phenotype distinguishes primitive lymphoid progenitors from pluripotent stem cells.

CD7 may cooperate with CD28 in Treg function, as CD28/CD7 double-deficient mice show reduced Treg numbers and suppressive activity.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted, tested by flow cytometry on human peripheral blood cells. Use 5 µL (0.25 µg) per test in 100 µL final volume.
Excitation/Emission: 405 nm / 436 nm (Violet laser).
Recommended Filter: 450/50 bandpass or equivalent.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Samples can be stored up to 3 days at 4°C in the dark using:

Clone-specific performance after fixation should be determined empirically.
