Product Specific Information
The 53-6.7 monoclonal antibody reacts with mouse CD8a, a 32–34 kDa cell surface receptor expressed as CD8αβ or CD8αα dimers.
- CD8αβ: majority of thymocytes and αβ TCR T cells
- CD8αα: γδ TCR T cells, intestinal IELs, dendritic cells
CD8 binds MHC class I and associates with protein tyrosine kinase p56^lck, playing a role in T cell development and activation.
Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes at ≤0.25 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize titration for best results.
Super Bright 436 is excited at 405 nm and emits at 436 nm. Recommended filter: 450/50 bandpass or equivalent. Ensure instrument compatibility.
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Target Information
CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein functioning as a co-receptor for MHC class I molecules.
- Expressed on T cells, NK cells, monocytes, and dendritic cells
- Exists as CD8αβ heterodimer or CD8αα homodimer
- Involved in signal transduction, T cell differentiation, and activation
Upon MHC-I/peptide ligation, CD8 recruits Lck kinase, leading to lymphokine production, motility, and CTL activation.
Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
