Product Specific Information

This GzA-3G8.5 monoclonal antibody reacts with mouse Granzyme A, the most abundantly expressed of the ten known granzyme serine proteases in mice. Granzymes are released from NK cells and cytotoxic T lymphocytes (CTLs) to induce target cell death by cleaving intracellular substrates, playing a key role in immune defense against viruses, tumors, and intracellular bacteria.

Granzyme A activates caspase-independent cell death pathways with mitochondrial and DNA damage, and may also contribute to inflammation by processing pro-IL-1β. It shares substrate specificity with Granzyme K, explaining minimal cytotoxicity reduction in Granzyme A-deficient CTLs.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Validated for intracellular staining and flow cytometry of mouse splenocytes (≤ 0.125 µg/test). Antibody should be titrated for optimal performance.

Fluorochrome Information:
PerCP-eFluor® 710 emits at 710 nm and is excited by the blue laser (488 nm). Recommended filter: 710/50 bandpass (695/40 acceptable alternative). Ensure instrument compatibility before use.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using:

Filtration: 0.2 µm post-manufacturing filtered


Target Information

Cytolytic T lymphocytes (CTLs) and natural killer (NK) cells recognize and lyse specific target cells presenting nonself antigens, typically peptides from intracellular pathogens. Granzyme A is a T cell- and NK cell-specific serine protease essential for target cell lysis and immune defense.
