Target Information

CD47 (integrin-associated protein, IAP) is a ubiquitously expressed transmembrane glycoprotein that interacts with several integrins and regulates their functions. Engagement of CD47 by soluble ligands or counter receptors modulates various signaling pathways, including activation of heterotrimeric G proteins. Functionally, CD47 contributes to intracellular calcium increase upon cell adhesion to extracellular matrix. It also acts as a receptor for thrombospondin’s C-terminal cell binding domain and may play roles in membrane transport and signal transduction.
CD47 is broadly distributed across tissues, reduced on Rh erythrocytes, and serves as a ligand for CD172a (SIRPα), an inhibitory receptor on macrophages, preventing phagocytosis of CD47-positive cells. The CD47-CD172a system influences cell migration, B cell adhesion, T cell activation, chondrocyte mechanotransduction, and neuronal development, being abundant in synapse-rich regions of brain and retina.
