Product Specific Information
The 5C3 monoclonal antibody reacts with human CD40, a 45–50 kDa type I transmembrane glycoprotein. CD40, a member of the TNFR family, is expressed by B lymphocytes, follicular dendritic cells, thymic epithelium, and a subset of peripheral T cells. It regulates B cell development and maturation by inducing Ig isotype-switching and, in combination with IL-4, protects B cells from apoptosis and promotes proliferation.
Interaction of CD40 with CD154 (gp39) on T cells is crucial for T–B cell communication and immune regulation. The 5C3 antibody is commonly used for B cell activation in in vitro functional assays.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells. Use 5 µL (0.6 µg) per test (100 µL final volume). Suitable for 10⁵–10⁸ cells/test.
Important Notes:
- NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for best compatibility.
- CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
- Add 5 µL per sample before mixing with cells.
- For single-color controls: 5 µL per 100 µL of cell sample (10³–10⁸ cells).
NovaFluor conjugates utilize Phiton™ technology, based on novel nucleic acid dye structures engineered for minimal spillover and optimized spectral performance.

Target Information
CD40 is a single-chain glycoprotein and a member of the TNF/NGF superfamily, homologous to the Hodgkin’s disease antigen CD30. It is present on all B cells (except plasma cells), epithelial cells, carcinomas, and lymphoid dendritic cells. CD40 is essential for immune and inflammatory responses, including:
- T cell-dependent immunoglobulin class switching
- Memory B cell development
- Germinal center formation
The AT-hook transcription factor AKNA regulates CD40 expression, mediating cell interactions. TNFR2 interacts with CD40 to mediate signal transduction. CD40–CD154 interaction is implicated in amyloid-beta-induced microglial activation, relevant to Alzheimer’s disease pathogenesis.
Two alternatively spliced transcript variants of CD40 are known.
CD40 dysfunction is associated with Type 3 Hyper-IgM immunodeficiency and CD40 ligand deficiency.
