Product Specific Information

The 5C3 monoclonal antibody reacts with human CD40, a 45–50 kDa type I transmembrane glycoprotein belonging to the TNFR family. It is expressed on B lymphocytes, follicular dendritic cells, thymic epithelium, and a subset of peripheral T cells.
CD40 plays a key role in B cell development, maturation, and survival by inducing Ig isotype-switching and preventing apoptosis in cooperation with IL-4. Interaction with CD154 (gp39) on T cells is essential for T-B cell crosstalk and immune regulation.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells. Use 5 µL (0.6 µg) per test (final volume 100 µL). Suitable for 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI.
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining. Add 5 µL per sample before mixing with cells.
• For single-color controls, use 5 µL of buffer per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology with nucleic acid dye structures engineered for minimal spillover and spread.

Target Information

CD40 is a single-chain glycoprotein belonging to the TNF/NGF superfamily and shares homology with CD30. It is expressed on all B cells except plasma cells, as well as some epithelial cells, carcinomas, and dendritic cells.
CD40 is essential for immune and inflammatory responses including immunoglobulin class switching, memory B cell development, and germinal center formation.
It interacts with adaptor protein TNFR2 and is implicated in amyloid-beta-induced microglial activation, relevant to Alzheimer’s disease pathogenesis.
Two alternatively spliced transcript variants encoding distinct isoforms have been reported.
Associated diseases include Type 3 Hyper-IgM immunodeficiency and CD40 ligand deficiency.
