Product Specific Information

The 5C3 monoclonal antibody reacts with human CD40, a 45–50 kDa type I transmembrane glycoprotein. CD40 is a TNFR family member expressed by B lymphocytes, follicular dendritic cells, thymic epithelium, and a subset of peripheral T cells.
It regulates B cell development and maturation, induces Ig isotype-switching, and promotes proliferation. Interaction with CD154 (gp39) on T cells is critical for immune regulation and costimulation.

5C3 is used for activation of B cells in in vitro functional assays.
Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells. Recommended use: 5 µL (0.6 µg) per test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test.

Compatibility:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
Recommended viability stains: Invitrogen LIVE/DEAD Fixable Dead Cell stains.

Buffer Usage:
CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
Add 5 µL per sample or per 100 µL cell sample (10³–10⁸ cells) for single-color controls.

NovaFluor conjugates use Phiton™ technology with engineered fluorescent signatures optimized for spillover and spread impacts.

Target Information

CD40 is a single-chain glycoprotein and member of the TNF/NGF superfamily.
Expressed on all B cells except plasma cells, and also found on epithelial cells, carcinomas, and lymphoid dendritic cells.
It mediates immune and inflammatory responses including:

• T cell-dependent immunoglobulin class switching
• Memory B cell development
• Germinal center formation

AKNA transcription factor regulates CD40 expression.
CD40 interaction with TNFR2 mediates signal transduction.
Its ligand interaction is implicated in amyloid-beta-induced microglial activation, relevant to Alzheimer’s disease.
Two alternatively spliced transcript variants encode distinct isoforms.
Associated diseases include Type 3 Hyper-IgM immunodeficiency and CD40 ligand deficiency.
