Product Specific Information

The OKT4 monoclonal antibody reacts with human CD4, a 59 kDa cell surface glycoprotein expressed by thymocytes, mature T-helper cells, and monocytes. CD4 acts as a receptor for HIV. The OKT4 antibody recognizes a distinct epitope from the RPA-T4 antibody and does not cross-block.

Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Applications Reported:

• Suitable for flow cytometric analysis.

Applications Tested:

• Tested on normal human peripheral blood cells.
• Use ≤5 µL (0.6 µg) per test (100 µL final volume).
• Recommended to titrate for optimal performance.
• Cell range: 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• For optimal staining, use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01).
• Add 5 µL of blocking buffer per sample before combining with cells.
• For single-color controls, use 5 µL per 100 µL cell sample containing 10³–10⁸ cells.

NovaFluor conjugates utilize Phiton™ technology with engineered fluorescent signatures to minimize spillover and spread.

Target Information

CD4 is a co-receptor for MHC class II molecules and HIV. It is expressed in T-helper cells, monocytes, macrophages, and dendritic cells. At the tissue level, CD4 is found in thymus, lymph nodes, tonsils, spleen, and certain regions of the brain and gut. It plays a role in T-cell activation via association with the TCR complex and tyrosine kinase Lck. CD4 may also mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts are known (RefSeq, July 2017).
