Product Specific Information

The eBioA1 monoclonal antibody reacts with human CD39, also known as ectonucleoside triphosphate diphosphohydrolase 1 (ENTPD1) or NTPDase. CD39 is an integral membrane protein with two transmembrane domains and exists as a homotetramer. It functions to remove toxic extracellular ATP by converting it to ADP or AMP, and works together with CD73 to hydrolyze ATP.

CD39 expression was initially identified on activated lymphocytes and is also found on subsets of T cells, B cells, dendritic cells, and weakly on monocytes and granulocytes. It is expressed on regulatory T cells (Treg), facilitating their entry into inflamed areas with high ATP levels. Expression on Foxp3+CD4+ cells ranges from 25–45%.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested with normal human peripheral blood cells at 5 µL (0.8 µg)/test.
A test is defined as staining a cell sample in a final volume of 100 µL. Recommended cell number: 10⁵–10⁸ cells/test. Optimize titration for best performance.

Compatibility Notes:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.

Blocking Buffer Guidance:
This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which contains improved formulation over CellBlox. Add 5 µL per sample to antibody cocktails before combining with cells. For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates are based on Phiton™ technology utilizing nucleic acid dye structures for engineered fluorescent signatures optimized for minimal spillover and spectral spread.

Target Information

CD39 is a 70–100 kD molecule expressed on peripheral blood B cells, monocytes, T cell clones, and weakly on granulocytes. It exhibits ecto-ATPase activity and is induced on T cells and increased on B cells as a late activation antigen.
