Product Specific Information

The OKT4 monoclonal antibody reacts with human CD4, a 59 kDa cell surface glycoprotein expressed by the majority of thymocytes, a subpopulation of mature T cells (T-helper cells), and at low levels on monocytes. CD4 is a receptor for HIV. The OKT4 antibody recognizes a different epitope than the RPA-T4 monoclonal antibody, and these antibodies do not cross-block each other.

Each product contains:

• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on normal human peripheral blood cells. Recommended ≤5 µL (1.0 µg) per test in 100 µL final volume. Cell number range: 10⁵–10⁸ cells/test.

Usage Notes:

• Carefully titrate for optimal performance.
• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):

• Improved formulation over CellBlox.
• Required for optimal staining with NovaFluor conjugates.
• Add 5 µL per sample to antibody cocktails or single-color controls (100 µL cell sample, 10³–10⁸ cells).

NovaFluor conjugates utilize Phiton technology with engineered fluorescent signatures optimized for minimal spillover and spread impacts.

Target Information

CD4 antigen is involved in recognition of MHC class II molecules and acts as a co-receptor for HIV. CD4 is primarily expressed in T-helper cells but also in monocytes, macrophages, and dendritic cells. Expression is observed in thymus, lymph nodes, tonsils, spleen, and certain non-lymphoid tissues such as brain and gut. CD4 contributes to T-cell activation and may mediate neuronal damage in infectious or immune diseases of the CNS. Multiple alternatively spliced transcripts identified (RefSeq, July 2017).
