Product Specific Information

Description:
The OKT4 monoclonal antibody reacts with human CD4, a 59 kDa cell surface glycoprotein expressed by most thymocytes, T-helper cells, and at low levels on monocytes. CD4 serves as a receptor for HIV. OKT4 recognizes a different epitope than RPA-T4 and does not cross-block binding.

Each product includes 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.

Applications Reported:
Suitable for flow cytometric analysis.

Applications Tested:
Tested on normal human peripheral blood cells. Recommended usage ≤5 µL (0.6 µg) per test (in 100 µL final volume). Optimize titration for best performance. Cell number: 10⁵–10⁸ cells/test.

Compatibility Notes:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

Blocking Buffer Usage:

• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
• Add 5 µL per sample (regardless of number of NovaFluors).
• For single-color controls: add 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates use Phiton™ technology with engineered nucleic acid dye structures for optimized fluorescence performance and minimal spectral spillover.

Target Information

CD4 antigen participates in MHC class II recognition and serves as a co-receptor for HIV. Expressed mainly on T-helper cells, monocytes, macrophages, and dendritic cells. Found in thymus, lymph nodes, tonsils, spleen, and certain non-lymphoid tissues. CD4 aids in early T-cell activation via association with the TCR complex and Lck kinase. Multiple splice variants exist (RefSeq, July 2017).
