Target Information

The androgen receptor (AR) is a member of the steroid-hormone receptor superfamily of nuclear receptors.
It is a ligand-activated transcription factor (>90 kDa) with three major functional domains:

  1. N-terminal domain
  2. DNA-binding domain
  3. Androgen-binding domain

Upon binding testosterone (T) or dihydrotestosterone (DHT), AR dissociates from accessory proteins, translocates into the nucleus, dimerizes, and activates transcription of androgen-responsive genes.

The AR signaling pathway is essential for development and function of male reproductive organs (prostate, epididymis) and also active in nonreproductive tissues (muscle, hair follicles, brain).
AR is a phosphoprotein that regulates MAP kinase signaling. Inhibition of MEK1/2 affects AR phosphorylation.
Abnormal AR signaling is associated with prostate cancer, Kennedy’s disease, male infertility, and complete androgen insensitivity (CAIS).
