Product Specific Information

The OKT3 monoclonal antibody reacts with an epitope on the epsilon-subunit within the human CD3 complex. It has potent immunosuppressive properties and has been used effectively in treatment of organ allograft rejection. CD3 is expressed by thymocytes and all mature T cells, and crosslinking of TCR initiates intracellular activation and proliferation pathways. Clones OKT3 and SK7 recognize different epitopes.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested for flow cytometry at 5 µL (0.5 µg) per test.
A test is defined as the amount of antibody that stains a cell sample in a final volume of 100 µL, with 10⁵–10⁸ cells/test.

Fluorochrome Information:
Brilliant Ultra Violet™ 805 (BUV805) emits at 797 nm and is designed for cytometers with a 355 nm UV laser. Ensure instrument compatibility.

Staining Buffer Recommendation:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (SB-4401) or Brilliant Stain Buffer (00-4409-75) to minimize non-specific interactions.

Light Sensitivity:
Protect from light to prevent photo-induced oxidation.

Fixation:
Samples can be stored in IC Fixation Buffer (00-8222) or 1-step Fix/Lyse Solution (00-5333) for up to 3 days at 4°C in the dark.
BUV805 is compatible with short-term methanol fixation but should not be stored in methanol-containing buffers for longer than one hour.

Target Information

The CD3 subunit complex transduces antigen-recognition signals into T cells and regulates TCR surface expression.
CD3 subunits (gamma, delta, epsilon, zeta) are encoded on chromosome 11 and associate with cytoplasmic signaling molecules via tyrosine-based motifs.
CD3 is present on 68–82% of peripheral blood lymphocytes and 65–85% of thymocytes, but not on B or NK cells.
Defects in CD3 genes are linked to CD3 immunodeficiency.
