Product Specific Information

The monoclonal antibody 2D3 reacts with CD47, also known as integrin-associated protein (IAP) and neurophilin.
CD47 is a glycosylated five-transmembrane protein involved in adhesion via interactions with SIRP (signal regulator protein) and integrins (CD51/CD61, CD41/CD61). It mediates bi-directional signaling that affects neural synaptic activity and macrophage phagocytosis. CD47 also functions as a receptor for thrombospondin.

CD47 is expressed in most hematopoietic cells (T, B cells, monocytes, platelets, erythrocytes) and non-hematopoietic tissues.

Each product includes:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications

  • Reported for use in flow cytometric analysis
  • Pre-titrated and tested on normal human peripheral blood cells
  • Recommended use: 5 µL (0.6 µg) per test
  • A test = staining of cell sample in 100 µL final volume (10⁵–10⁸ cells/test)

Important Notes

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for compatibility.
  • CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
    • Add 5 µL per sample before combining with cells.
    • For single-color controls: add 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

CD47 (Integrin-associated protein, IAP) is a ubiquitous transmembrane glycoprotein that interacts with integrins and regulates signaling pathways, including G protein activation and calcium mobilization upon cell adhesion.
It serves as a receptor for thrombospondin and modulates membrane transport, signal transduction, and immune functions.
CD47 acts as a ligand for CD172a (SIRPα), preventing phagocytosis of CD47-positive cells and influencing cell migration, B cell adhesion, T cell activation, and neuronal development.
