Product Specific Information

The monoclonal antibody 2D3 reacts to CD47, also known as integrin-associated protein (IAP) and neurophilin.
CD47 is a glycosylated five-transmembrane protein involved in adhesion through interactions with SIRP and integrins (CD51/CD61, CD41/CD61). It mediates bidirectional signaling affecting neural synaptic activity and macrophage phagocytosis. CD47 also serves as a receptor for thrombospondin and is expressed in most hematopoietic and non-hematopoietic cells.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for flow cytometry using normal human peripheral blood cells.
Use at 5 µL (0.6 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Compatibility Notes:
NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI.
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):
Required for optimal staining with NovaFluor conjugates.
Add 5 µL per sample before combining antibodies with cells.
For single-color controls, use 5 µL per 100 µL cell sample.

NovaFluor Conjugates:
Based on Phiton™ technology utilizing nucleic acid dye structures for engineered fluorescence minimizing spillover and spread.

Excitation: 494 nm
Emission: 585 nm
Laser: 488 nm (Blue)

A ubiquitously expressed transmembrane glycoprotein interacting with integrins and modulating signaling pathways.
Functions include:

• Regulation of intracellular calcium upon adhesion
• Receptor for thrombospondin
• Ligand for CD172a (SIRP alpha), inhibiting macrophage phagocytosis
• Involved in cell migration, B cell adhesion, T cell activation, and neuronal development
  Highly expressed in brain and retina, reduced on Rh erythrocytes.
