Product Specific Information

The monoclonal antibody 2D3 reacts with CD47, also known as integrin-associated protein (IAP) and neurophilin.
CD47 is a glycosylated five-transmembrane protein involved in adhesion via interactions with SIRP (signal regulator protein) and beta3 integrins (CD51/CD61 and CD41/CD61). These interactions mediate bi-directional signaling affecting neural synaptic activity and macrophage phagocytosis. CD47 also serves as a receptor for thrombospondin and is expressed in most hematopoietic and non-hematopoietic cells.

Each product includes:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

  • Suitable for flow cytometric analysis

Applications Tested

  • Pre-titrated and tested on normal human peripheral blood cells
  • Recommended use: 5 µL (0.6 µg) per test
  • A test is defined as staining in a 100 µL final volume
  • Cell number: 10⁵–10⁸ cells/test (empirically determined)

Important Notes

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI.
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell Stains instead.
  • Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining with NovaFluor conjugates.
  • Add 5 µL per sample to antibody cocktails before combining with cells.
  • For single-color controls: use 5 µL of blocking buffer per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, employing novel nucleic acid dye structures engineered to minimize spillover and spread in multicolor flow cytometry.

Target Information

CD47 (integrin-associated protein, IAP) is a ubiquitously expressed transmembrane glycoprotein that interacts with several integrins to regulate their functions.
Engagement with ligands or counter receptors modulates signaling pathways, including activation of heterotrimeric G proteins and calcium signaling upon cell adhesion.
CD47 also serves as a receptor for the C-terminal domain of thrombospondin, playing roles in membrane transport, signal transduction, and immune regulation.
The CD47–CD172a (SIRPα) interaction inhibits macrophage-mediated phagocytosis of CD47-positive cells and influences cell migration, adhesion, and neuronal development.
