Target Information
Furin is a membrane-associated, calcium-dependent serine protease belonging to the subtilisin-like prohormone convertase (PC) family. These enzymes cleave prohormones and neuropeptide precursors, as well as various cellular, viral, and bacterial proteins transported via the secretory pathway.
Furin shares structural similarities with other PC family members, including:
- ~10 kDa amino-terminal proregion
- ~55 kDa conserved catalytic domain
- Variable-length carboxyl-terminal domain
Furin becomes active after proregion cleavage and is the only PC with a transmembrane domain. Cleavage occurs at the carboxyl-terminus of the furin consensus sequence RX(K/R)R.
The acidic peptide sequence (C771PSDSEEDEG780) localizes furin to the trans-Golgi network, and serine phosphorylation modulates routing.
The tetrapeptide sequence (Y759KGL762) directs internalization from the cell surface.
