Target Information

Proteolytic degradation is critical to maintaining appropriate levels of short-lived and regulatory proteins, including those involved in cellular metabolism, heat shock response, antigen presentation, and cell cycle regulation.
The ubiquitin-proteasome pathway deconstructs most cytosolic and nuclear proteins, while others are degraded via the vacuolar pathway involving endosomes, lysosomes, and the endoplasmic reticulum.
The 26S proteasome is an ATP-dependent, multisubunit molecular complex (~2.5 MDa) composed of a 20S core and 19S regulatory subunits. The 19S subunit recognizes ubiquitinated proteins and facilitates their unfolding and translocation into the 20S core.
Defects in this pathway are associated with genetic diseases such as cystic fibrosis, Angelman’s syndrome, and Liddle syndrome.
