Apoptotic signals are often triggered by cell surface death receptors through protein–protein interactions mediated by conserved domains such as the death effector domain (DED).
DEDD2 is a novel DED-containing protein identified to play a role in apoptosis regulation. Overexpression of DEDD2 induces moderate apoptosis and sensitizes cells to Fas, TRAIL, and FADD-induced apoptosis.
DEDD2 binds caspase-8 and -10, as well as FLIP, but not FADD. It translocates from the cytosol to the nucleus upon apoptosis induction, suggesting a role in targeting caspase-8 to the nucleus and mediating death receptor-induced apoptosis.
Two alternatively spliced transcript variants encoding distinct isoforms have been identified.
