Product Specific Information

Description:
The 93 monoclonal antibody binds to an epitope shared by mouse CD16 and CD32.
CD16 (FcγRIII) and CD32 (FcγRII) are low-affinity receptors for the Fc portion of mouse IgG, expressed on B cells, monocytes/macrophages, NK cells, and neutrophils.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Mouse splenocytes, ≤1.0 µg/test (100 µL final volume, 10⁵–10⁸ cells/test).
Optimize antibody titration for best performance.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for compatibility.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
  • Add 5 µL per sample (regardless of the number of NovaFluor dyes).
  • For single-color controls: 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates use Phiton™ technology, employing nucleic acid dye structures engineered for optimal fluorescence and minimal spectral spillover.

Target Information

CD16 (FcγRIII):

• 50–65 kDa cell surface molecule
• Expressed as transmembrane (NK, some T cells) and GPI-linked (granulocytes) forms
• Mediates antibody-dependent cell-mediated cytotoxicity (ADCC)

CD32 (FcγRII):

• 40 kDa glycoprotein
• Low-affinity IgG receptor
• Functions: endocytosis, secretion activation, cytotoxicity, immunomodulation
• Expressed on B cells, monocytes, granulocytes, platelets
• Involved in phagocytosis of immune complexes and antibody production regulation
• Gene variations may increase susceptibility to systemic lupus erythematosus (SLE)
• Multiple transcript variants exist
