Product Specific Information

Description:
The 93 monoclonal antibody reacts with an epitope shared by mouse CD16 and CD32. CD16 (Fc gamma III Receptor) and CD32 (Fc gamma II Receptor) are low affinity receptors for the mouse IgG Fc portion and are expressed by B cells, monocyte/macrophages, NK cells, and neutrophils.

Each product contains 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.

Applications Reported:
Reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤1.0 µg per test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Optimize antibody titration for best performance.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
• Add 5 µL per sample to antibody cocktails before combining with cells.
• For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Based on Phiton™ technology utilizing novel nucleic acid dye structures for engineered fluorescent signatures with minimized spillover and spread effects.

Target Information

CD16:
A 50–65 kDa surface molecule existing in transmembrane (NK cells, some T cells) and phosphatidylinositol-linked (granulocytes) forms. Functions as a low-affinity IgG receptor (FcR III) mediating antibody-dependent cell-mediated cytotoxicity (ADCC) by NK cells.

CD32:
A 40 kDa glycoprotein functioning as a low-affinity IgG receptor (Fc gamma RII), involved in endocytosis, activation of secretion, cytotoxicity, and immunomodulation. Expressed by B cells, monocytes, granulocytes, and platelets. Plays roles in immune complex phagocytosis and regulation of antibody production. Variants may influence susceptibility to systemic lupus erythematosus (SLE).
